2、養(yǎng)因子(glialcellline—derivedneurotrophicfactor,GDNF)對其表達(dá)的影響。方法:實(shí)驗(yàn)1:雄性SD大鼠隨機(jī)分為假手術(shù)組(sham組)和坐骨神經(jīng)結(jié)扎組(CCI組)。CCI組又分為術(shù)后ld(D1)、3d(D3)、7d(D7)和14d(D14)組。實(shí)驗(yàn)2:雄性SD大鼠隨機(jī)分為:sham組、D14組、D14+GDNF組和D14+PBS組。兩實(shí)驗(yàn)分別于實(shí)驗(yàn)前及CCI術(shù)后第1、3、7、14d測定熱縮足潛伏期(thermalwithdrawallatency,TwL)及機(jī)械縮足閾值(mechanicalwithdrawa
3、lthreshold,MWT),處死后取其L4~k脊髓背角,采用Westemblot方法測定GAD65蛋白表達(dá)情況。結(jié)果:與sham組相比,CCI組大鼠TwL及MwT均降低,GAD65表達(dá)水平于術(shù)后3d開始降低,一直持續(xù)到術(shù)后14d;與D14組和D14+PBS組相比,D14+GDNF組大鼠TwL及MWT均升高,GAD65表達(dá)水平升高。結(jié)論:神經(jīng)病理性疼痛的發(fā)生機(jī)制可能和大鼠脊髓背角內(nèi)GAD65表達(dá)降低相關(guān),而GDNF對其鎮(zhèn)痛作用可能部分是通過增加脊髓GAD65的表達(dá)實(shí)現(xiàn)的。關(guān)鍵詞膠質(zhì)細(xì)胞源性神經(jīng)營養(yǎng)因子;神經(jīng)病理性疼痛;谷氨酸脫羧酶65EI’F
