1、云南紅豆杉紫杉醇含量變異及其相關(guān)的RAPD分子標(biāo)記第45卷第7期2009年7月林業(yè)科學(xué)SCIENTIASILVAESINICAEVo1.45.No.7Ju1.,2009云南紅豆杉紫杉醇含量變異及其相關(guān)的RAPD分子標(biāo)記蘇建榮繆迎春張志鈞(中國林業(yè)科學(xué)研究院資源昆蟲研究所昆明650224)摘要:從5個(gè)云南紅豆杉群體中分別選取樹齡與生長狀況基本一致的植株各10個(gè),按統(tǒng)一的取樣標(biāo)準(zhǔn)采集樹皮,小枝和針葉樣品測定紫杉醇含量,分析其變異分布式樣;同時(shí)對(duì)所有采樣植株進(jìn)行RAPD分析,開展RAPD標(biāo)記與不同器官紫杉醇含量的關(guān)聯(lián)分析.結(jié)果表明:不
2、同云南紅豆杉群體問樹皮,小枝和針葉的紫杉醇含量均差異顯著(P<0.05),樹皮,小枝和針葉紫杉醇含量在群體間的變異分別占其總變異的16.22%,33.90%和45.81%.通過RAPD標(biāo)記與紫杉醇含量的關(guān)聯(lián)分析,分別篩選出與云南紅豆杉樹皮,小枝和針葉紫杉醇含量相關(guān)聯(lián)的位點(diǎn)11,l3,59個(gè),其貢獻(xiàn)率依次為7.791%一11.509%,7.853%24.457%,7.858%~26.729%;同時(shí)篩選出了可指示樹皮,小枝和針葉低紫杉醇含量的特異標(biāo)記.云南紅豆杉的遺傳改良應(yīng)根據(jù)不同器官紫杉醇含量變異的分布特點(diǎn)結(jié)合葉用,枝用和皮
