1、大豆蚜捕食性天敵捕食行為的COI基因標記檢測宋新元,叢斌,錢海濤,董輝(沈陽農(nóng)業(yè)大學植物保護學院,沈陽110161)摘要:【目的】探索大豆田捕食性天敵對大豆蚜的捕食作用?!痉椒ā扛鶕?jù)基因庫NCBI中的一段大豆蚜細胞色素氧化酶I(COI)的基因序列(登錄號為AY842503),設計了2對大豆蚜特異引物A和B,其擴增片段大小分別約為197和253bp,應用DNA標記方法檢測天敵對大豆蚜的捕食行為?!窘Y(jié)果】種特異性檢驗表明,所設計引物只對大豆蚜DNA具有擴增效果,對與其同域發(fā)生的其它種類不具擴增作用;室內(nèi)以引物對A與B分別檢測喂食單頭大豆蚜的異色瓢
2、蟲成蟲和大草蛉成蟲腹內(nèi)靶標食物的衰變情況,結(jié)果表明,異色瓢蟲成蟲陽性比率的半衰期分別為喂食后3.371h和2.814h,大草蛉成蟲分別為喂食后1.312h和1.032h;以A作為引物對田間捕食性天敵進行檢測,結(jié)果表明,在所檢測的7類群14種捕食性天敵中,5種不同種類或蟲態(tài)的天敵對大豆蚜的捕食作用檢出率高于50%,從高到低依次為異色瓢蟲幼蟲、草蛉成蟲、東亞小花蝽若蟲、異色瓢蟲成蟲、東亞小花蝽成蟲;此外,田間檢測表明,捕食性天敵的陽性比率與大豆蚜的種群密度呈顯著正相關(guān)?!窘Y(jié)論】DNA標記技術(shù)是探索捕食行為的有效方法之一。關(guān)鍵詞:大豆蚜;細胞色素氧
