1、重慶醫(yī)科大學碩士學位論文萊菔硫烷對原代培養(yǎng)神經(jīng)元氧糖剝奪/復氧損傷的保護作用及機制研究姓名:吳雪梅申請學位級別:碩士專業(yè):病理學與病理生理學指導教師:趙涌201205重慶醫(yī)科大學碩士研究生學位論文萊菔硫烷對原代培養(yǎng)神經(jīng)元氧糖剝奪/復氧損傷的保護作用及機制研究摘要目的:探討萊菔硫烷(SFN)對氧糖剝奪(OGD)/復氧(R)損傷的皮質(zhì)神經(jīng)元凋亡的影響并探討其與P13K/AKT通路的關(guān)系。方法:(1)新生1d的SD乳大鼠,皮質(zhì)神經(jīng)元原代培養(yǎng)5-6d后,細胞被隨機分為正常對照組、OGD組、SFN組,SFN+LY組及LY組,建立皮質(zhì)神經(jīng)元的OGD模型。(2)1h后復氧,
2、模擬再灌注模型,OGD/R期間給予O.1、0.2、O.5、l、2.5、5pmol/LSFN,P13K抑制劑LY294002(10pmol/L)在OGD/R期間與SFN或不與SFN同時加入神經(jīng)元。(3)復氧24h后,采用四甲基偶氮唑鹽(MTT)法測定神經(jīng)元的存活率,Hoechst33258/PI聯(lián)合染色法測定細胞損傷程度,免疫熒光和Westernblot觀察神經(jīng)元OGD瓜后Bcl一2、cleavedcaspase-3、Akt和p-Akt的表達。結(jié)果:(1)MTT結(jié)果顯示在SFN處理濃度在1}tmol/L時,可顯著增加神國家自然科學基合咨助(No.30900457
