2、汐、姐翻:通過對DahlS高血壓大鼠卵巢細胞凋亡及相關因素的研究,以期闡明高血壓時卵巢細胞凋亡的特征及機制并尋找防治高血壓改善卵巢功能的途徑。,—喃零去;以日本引進DahlS高血壓大鼠為研究對象。(1)選用生后5周的DahlS雌性大鼠隨機分為對照組、實驗組,分別投予正常、8%食鹽飼料,于1l、13、15周齡取材。(2)選用生后5周的DahlS雌性大鼠投予8%食鹽飼料,予1l周齡時隨機分為二組,經鼠尾靜脈注射bFGF為給藥組;注射
3、生理鹽水為未給藥組,治療結束后取材。應用光鏡、電鏡,TUNEL及SP染色,流式細胞儀,放射免疫分析,生化等技術,研究在高血壓因素下性激素、Bcl習、NO及iNOS、bFGF等相關因素與卵巢細胞凋亡的關系。,,矗結果:(1)光鏡下觀察實驗組隨高血壓病程延長、病情加重,生長卵泡中顆粒細胞排列不整齊,松散,閉鎖卵泡數目增多,黃體數目減少。(2)TUNEL染色可見凋亡在生長期各級卵泡的顆粒細胞中均有發(fā)生,閉鎖卵泡顆粒細胞凋亡最顯著,黃體細胞未見TUNEL陽性細胞。(3)電鏡下觀察實驗組隨高血壓病程延長,病情加重,可見凋亡的顆粒細胞
4、及黃體細胞增多,其特征為核周間隙增寬,核固縮,染色質邊集,凝聚,線粒體蜻模糊紊亂,腔內出現(xiàn)高電子密度異常顆粒,滑面內質網減少,類脂呈板層狀改變的數目增多。(4)流式細胞儀檢測:隨高血壓病情加重,卵巢細胞凋亡率逐漸上升,13、15周齡實驗組凋亡率分別為(6.42±1.06)%、(6.67±2.11)%均明顯高于對照組,且P5、09±0.32)nmol/k、(2.53±0.74)IU/L較對照組分別為(3.89±2.24)nmol/L和(0.64±0.31)IU/L,兩者比較差別顯著(P<0.01)。(6)給予注射bFGF治療后,流式細胞儀測定卵巢組織bFGF表達率(84.38±3.67)%較未給藥組(67.46±5.32)%顯著增高(P6、7.02±5.68),兩者比較差異顯著且P7、rchonApoptosisOfOvarianCel8、inDahlSandltsReIatedFactorHypertensiveRatAbstractObjective:Toelucidatethecharacterandmechanismofhypertensionleadingapoptosisofovaryandfindthewayofamelioratingthefunctionofovarybyreducingbloodpressure,theapoptosisanditsrelatedfactorsofova9、riancellinDahlShypentersionratswereresearched.Methods:DahlShytertensiveratsimportedfromJapanwereselectedassubjects.(1)DahlSfemaleratswereselectedat5postratalweekandderidedintoexperimentedgroupsandcontrolsrandomly.thentheywerefedwith8%saltandnormalforagerespectivel10、y.Materialsweregatheredat11,13,15weekold.(2)DahlSfemaleratswereselectedat5postratalweekandfedwith8%saltforage.thenweredividedintotwosubgroupsrandomly,ra
