1、膽鹽輸出泵在膽道梗阻再通大鼠肝組織中的表達(dá)及意義作者:祝建勇別平陳應(yīng)果馮春林唐春吳喬【摘要】目的探討膽鹽輸出泵(BSEP)在膽道梗阻再通大鼠肝組織中的表達(dá)及意義。方法將54只Wistar大鼠隨機(jī)分為假手術(shù)(Sham)組、膽總管結(jié)扎(CBDL)組、膽道再通(BR)組,測定各時(shí)相點(diǎn)血清丙氨酸氨基轉(zhuǎn)移酶(ALT)、總膽汁酸(TBA)以及腫瘤壞死因子(TNFct)的水平,采用RTPCR方法和二步法免疫組化染色分別在基因轉(zhuǎn)錄、蛋白表達(dá)及肝細(xì)胞膜定位上檢測BSEP在大鼠肝組織中的表達(dá)情況,運(yùn)用Mias99病理圖像分析系統(tǒng)半定量檢測BSEP的平均積分光密度仃0D),與TBA和TNFoc作相關(guān)分
2、析。結(jié)果CBDL術(shù)后大鼠血清ALT,TBA及TNFa顯著增高(P<0.05);BR術(shù)后上述指標(biāo)逐步好轉(zhuǎn),7d時(shí)仍顯著高于Sham組水平(P<0.05)oBSEPmRNA水平隨著梗阻時(shí)間的延長明顯降低(P<0.05),CBDL術(shù)后14d僅微弱表達(dá),梗阻解除后緩慢增高;BR術(shù)后7d仍未恢復(fù)至Sham組水平。免疫組化染色顯示BSEP表達(dá)定位于肝細(xì)胞膜上,變化規(guī)律與前述一致,并且與TBA和TNFex呈顯著負(fù)相關(guān)(rl=-0.9257,r2=-0.8536,P<0.05)。結(jié)論膽道梗阻再通大鼠模型中膽鹽的代謝與BSEP水平密切相關(guān),而TNFa可能是影響B(tài)SEP變化的
