3、me.Deconstructingthegenomeintomanageablefragmentsaffordstheplasticitynecessarytorapidlyintroducetargetedgeneticchangesinparallelandassortmutatedfragmentswhilemaximizinggenomestabilityovertime.Inthisprotocolwedescribeawell-developedreversegeneticplatformstrategyforCoVsthatiscomprisedofpartition
4、ingtheviralgenomeinto5–7independentDNAfragments(dependingontheCoVgenome),eachsubclonedintoaplasmidforincreasedstabilityandeaseofgeneticmanipulationandamplification.CoronavirusgenomesareconvenientlypartitionedbyintroducingtypeIISorIIGrestrictionenzymerecognitionsitesthatconferdirectionalcloning
