2、立快速檢測H5亞型禽流感病毒(AⅣ)的方法,本研究根據(jù)GenBank中登錄的H5亞型AIVHA基因序列,設(shè)計了一套特異識別HA基因序列中6個不同區(qū)段的環(huán)介導(dǎo)恒等溫擴增(LAMP)引物,建立了基于LAMP技術(shù)檢測H5亞型AIV的檢測方法。結(jié)果表明,該方法對H5AIVRNA的最小檢測限為0.1pfu,靈敏度高于普通一步法RT.PCR10倍;擴增反應(yīng)可以在30min內(nèi)完成;在反應(yīng)體系中添加鈣黃綠素(FD)熒光染料后,可以通過肉眼觀察有無熒光直接判定結(jié)果。靈敏性及特異性試驗表明,該方法靈敏度高、特異性好,能夠
