1、·674·寧夏醫(yī)學(xué)雜志2014年8月第36卷第8期NingxiaMedJ,Aug.2014Vol36,No.Doi:10.13621/j.1001—5949.2014.08.0674·論著·入—鼠電點(diǎn)燃島葉致癇模型建立后神經(jīng)肽Ghrelin的表達(dá)劉諍,王峰,宋子木,劉陽,高攀,李揚(yáng),孫濤[摘要]目的電點(diǎn)燃大鼠島葉建立致癇模型,檢測致癇大鼠神經(jīng)肽Ghrelin變化特征,并探討其意義。方法將雄性SD大鼠用數(shù)字表法隨機(jī)分為空白對照組、假手術(shù)組和點(diǎn)燃組,又將點(diǎn)燃組分為點(diǎn)燃后1、3、7d和10d4個(gè)亞組。立體定向下將電極植人大鼠島葉
2、,建立致癇模型,觀察大鼠行為學(xué),監(jiān)測大鼠腦電情況;Nissl染色觀察海馬區(qū)神經(jīng)元變化情況,用放射免疫法(RIA)和免疫熒光法分別檢測血清和下丘腦中Ghrelin的表達(dá)水平。結(jié)果點(diǎn)燃后各亞組血清Ghrelin表達(dá)水平均降低,差異有統(tǒng)計(jì)學(xué)意義(P<0.05);下丘腦Ghrelin表達(dá)也明顯減少,差異有統(tǒng)計(jì)學(xué)意義(P<0.05);各時(shí)間亞組間比較差異無統(tǒng)計(jì)學(xué)意義(P>0.05)。結(jié)論電刺激島葉可建立穩(wěn)定的點(diǎn)燃模型,癲癇發(fā)作后Ghrelin水平明顯降低,血清Ghrelin水平可以反映下丘腦中相應(yīng)神經(jīng)肽的變化趨勢。[關(guān)鍵詞]Ghre
