1、2O48實用醫(yī)學雜志2014年第30卷第13期右美托咪定對大鼠胚胎神經(jīng)干細胞增殖及分化的影響李紅英于衛(wèi)張建杏周國斌摘要目的:探討右美托咪定(dexmedetomidine,DEX)對大鼠胚胎神經(jīng)干細胞(NSCs)增殖及分化的影響。方法:從孕14—16dSD大鼠胚胎中分離NSCs并原代培養(yǎng)及Nestin鑒定。實驗分為對照組、低濃度及高濃度DEX組,培養(yǎng)液中分別加入終濃度為0、1、10ng/mLDEX。M1Tr法檢測NSCs活性;5一溴脫氧尿(Brdu)法觀察檢測NSCs增殖;Westernblot法檢測增殖期N
2、SCs中ERK1/2磷酸化:免疫熒光法標記神經(jīng)元細胞、星形膠質(zhì)細胞、少突膠質(zhì)細胞和神經(jīng)元干細胞觀察細胞分化。結(jié)果:分離培養(yǎng)的神經(jīng)干細胞95%以上呈Nestin陽性;低濃度DEX組M1Tr吸光值、Brdu陽性細胞及EKR1/2磷酸化水平較對照組明顯增加(P<0.O5)。而高濃度DEX組較對照組及低濃度DEX組明顯降低(P<0.05);高、低濃度DEX組中NeuN、GFAP及MBP陽性細胞數(shù)較對照組明顯增加(P<0.05),Nestin陽性細胞則顯著降低(P<0.05)。而高、低濃度DEX組間無明顯差異(P>0.
