1、中華麻醉學(xué)雜志2014年7月第34卷第7期ChinJAnesthesiol,July2014,Vo1.34,No.7873·重癥醫(yī)學(xué)P物質(zhì)對新生大鼠心肌細(xì)胞缺氧復(fù)氧損傷的影響趙鑫陳璐郭政【摘要】目的探討P物質(zhì)對新生大鼠心肌細(xì)胞缺氧復(fù)氧損傷的影響。方法新生sD大鼠,1~3日齡,處死后取心室肌組織,分離心肌細(xì)胞,接種于6孔培養(yǎng)板中,培養(yǎng)72h。采用隨機(jī)數(shù)字表法,將心肌細(xì)胞分為4組(n=3):對照組(C組)常規(guī)培養(yǎng)6h;缺氧復(fù)氧組(A/R組)進(jìn)行缺氧3h,復(fù)氧2h;P物質(zhì)組(sP組)給予終濃度為10一mol/L的P物質(zhì)孵育lh,行缺氧3h,復(fù)氧2h;P物質(zhì)+D.
2、sP組(sP+D.SP組)給予終濃度為10。mol/L的P物質(zhì)和終濃度為10mol/L的速激肽1型受體拮抗劑D.SP孵育1h,行缺氧3h,復(fù)氧2h。于復(fù)氧結(jié)束時(shí),測定心肌細(xì)胞凋亡率和乳酸脫氫酶(LDH)活性。結(jié)果與c組比較,A/R組、sP組和sP+D—sP組細(xì)胞凋率和LDH活性升高(P<0.01);與A/R組比較,sP組和sP+D.sP組細(xì)胞凋亡率和LDH活性降低(P<0.01);與sP組比較,sP+D—SP組細(xì)胞凋亡率和LDH活性升高(P<0.01)。結(jié)論P(yáng)物質(zhì)可減輕新生大鼠心肌細(xì)胞缺氧復(fù)氧損傷,其機(jī)制與激活速激肽1型受體有關(guān)。【關(guān)鍵詞】P物質(zhì);肌細(xì)胞,
