1、第31卷第3期海洋環(huán)境科學(xué)Vo1.31.No.32012年6月MARINEENVIRONMENTALSCIENCEJune2012氮、磷限制對(duì)塔瑪亞歷山大藻細(xì)胞周期及毒素含量和組成的影響高越,林琳,王大志(廈門大學(xué)環(huán)境科學(xué)研究中心近海海洋環(huán)境科學(xué)國家重點(diǎn)實(shí)驗(yàn)室,福建廈門361005)摘要:優(yōu)化了塔瑪亞歷山大藻AlexandriumtamarenseC101細(xì)胞同步化方法,研究了N、P限制對(duì)A.tamaremeC101細(xì)胞周期及毒素含量和組成變化的影響。結(jié)果表明,采用前期馴化、細(xì)胞沉降分離和光限制誘導(dǎo)相結(jié)合的同步化方法,細(xì)胞同步率提高了約14%。N、P限制條件下,細(xì)胞分裂受抑制
2、,分別停滯于G2/M期和G1期。營養(yǎng)鹽對(duì)毒素合成及組成影響比較明顯:N限制下單細(xì)胞毒素含量降低,P限制下單細(xì)胞毒素含量升高。在正常營養(yǎng)和P限制條件下,cll2占毒素總量的98%以上,GTX2.3占總毒素量的2%左右;N限制條件下,Ci,2所占比例下降到90%左右,而GTX2.3比例增長(zhǎng)到約10%。根據(jù)c1.2和GTX2在一個(gè)細(xì)胞周期內(nèi)的變化規(guī)律推測(cè),毒素的合成受光誘導(dǎo)并在細(xì)胞周期G1期的特定時(shí)間內(nèi)合成,細(xì)胞可能先合成GTX23毒素,后被轉(zhuǎn)化為c12毒素。關(guān)鍵詞:塔瑪亞歷山大藻;營養(yǎng)鹽限制;細(xì)胞周期;麻痹性毒素;毒素合成中圖分類號(hào):Q945.78文獻(xiàn)標(biāo)識(shí)碼:A文章編號(hào):1007
