2、OV3凋亡及生長抑制作用。方法噻唑藍法檢測Smac小分子compound3單獨應用組、順鉑單獨應用組及二者聯(lián)合應用組對卵巢癌細胞SKOV3的生長抑制率、存活率的影響;并繪制生長曲線。AnnexinV/PI雙標流式細胞術(shù)檢測Smac小分子compound3單獨應用組、順鉑單獨應用組及二者聯(lián)合應用組對SKOV3凋亡率的影響。Western印跡檢測Smac小分子compound3單獨應用組、順鉑單獨應用組及二者聯(lián)合應用組對SKOV3的x連鎖凋亡抑制蛋白(X—linkedinhibitorofapoptosisprotein,XIAP)、半胱氨酸一天冬氨酸蛋白
3、酶一3(caspase一3)的表達影響。結(jié)果Smac小分子compound3單獨應用組、順鉑單獨應用組及二者聯(lián)合應用組12、24、48hSKOV3生長受到抑制,且呈時間依賴性。二者聯(lián)合應用組與順鉑單獨應用組比較生長抑制率明顯升高。二者聯(lián)合應用組的凋亡率高于順鉑單獨應用組及Smac小分子compound3單獨應用組。Smac小分子compound3單獨應用組及二者聯(lián)合應用組SKOV3的XIAP表達降低,二者聯(lián)合應用組的caspase一3的激活片段表達明顯高于其他兩組。結(jié)論Smac小分子compound3能夠調(diào)節(jié)順鉑對SKOV3生長抑制作用,促進順鉑對卵巢
