2、腹中線開腹后將H22細胞接種到模型組小鼠肝臟實質(zhì)內(nèi),對照組注射生理鹽水。于注射后第l5天行剖腹探查,觀察各組成瘤率及腫瘤體積,并進行腫瘤病理學(xué)分析。結(jié)果各組小鼠的存活率均為100%。模型組I腫瘤移植成功率為100%,腫瘤體積平均為(65.214-5.25)mm。模型組II的腫瘤移植成功率為53.3%,腫瘤體積平均為(23.464-3.47)mm。模型組I的腫瘤平均體積明顯大于模型組II(P<0.05)。模型I組和II組病理檢查均證實為原位肝細胞瘤。結(jié)論成功建立了PLCe敲基因小鼠移植性肝癌動物模型,驗證了PLCe敲基因小鼠具有抑制肝腫瘤生長的特性。【關(guān)鍵詞】基因敲除;PLC8;移植性肝癌;
