1、通竅活血湯改善擬血管性癡呆大鼠學(xué)習(xí)記憶作用的研究葛朝亮,汪寧。(1.安徽醫(yī)科大學(xué)第一附屬醫(yī)院藥劑科,合肥230022;2.安徽中醫(yī)學(xué)院,安徽省中藥研究與開發(fā)重點實驗室,合肥230038)摘要:目的研究通竅活血湯對擬血管性癡呆(vasculardementia,VD)大鼠學(xué)習(xí)記憶作用及腦皮質(zhì)中乙酰膽堿含量的影響,探討通竅活血湯對擬血管性癡呆大鼠的作用機制。方法采用頸總動脈(CCA)注射混合血栓誘導(dǎo)劑法制作擬血管性癡呆大鼠模型。通過八臂迷宮檢測大鼠工作記憶錯誤次數(shù)、參考記憶錯誤;光鏡(HE染色)觀察擬血管性癡呆大鼠腦組織海馬CA1區(qū)病理形態(tài)的改變;酶聯(lián)免疫吸附試驗(ELISA)方法
2、檢測擬血管性癡呆大鼠腦皮質(zhì)中乙酰膽堿的含量。結(jié)果通竅活血湯高、中劑量能顯著減少擬血管性癡呆大鼠的工作記憶錯誤次數(shù)、參考記憶錯誤次數(shù)(P<0.O1);明顯改善擬血管性癡呆大鼠腦組織海馬CA1區(qū)病理形態(tài)學(xué)異常;顯著提高擬血管性癡呆大鼠腦皮質(zhì)中乙酰膽堿的含量(P<0.01)。結(jié)論通竅活血湯能明顯改善擬血管性癡呆大鼠的學(xué)習(xí)、記憶能力。其作用機制可能與改善擬血管性癡呆大鼠腦組織海馬CA1區(qū)的椎體細(xì)胞的病理形態(tài)的異常,提高擬血管性癡呆大鼠腦皮質(zhì)中乙酰膽堿的含量有關(guān)。關(guān)鍵詞:通竅活血湯;血管性癡呆;海馬CA1區(qū);乙酰膽堿中圖分類號:R965文獻標(biāo)志碼:A文章編號:1001—2494(2012
