4、ormelatonin-replacementrats(P<0.01).Hoelatoninreplacementtherapyreversedtheaboveparameters(P<0.01),-operatedrats(P>0.05).ConclusionThedataforthefirsttimedemonstratethatpinealectomyreducesthelevelofendogenousmelatonin,andimpairsthelearmingandmemoryprocessesascellsintheSVZ,elatoninreplacementthera
5、pycanreversethesechanges,indicatingthatmelatoninplaysanimportantroleinmaitainingthenormallearningandmemoryprocessesasentingtheneurogenesisinSVZ;andthatmelatoninmaypromotetheneurogenesisinSVZviaactivationofmelatoninreceptorsinbothneuralstemcellsandastrocytes,therebyenhancingolfactarymemory.哺乳動物側腦
6、室室管膜下區(qū)(subventricularzone,SVZ)是腦內終生存在神經(jīng)發(fā)生(neurogenesis)的一個主要部位,由此產(chǎn)生的神經(jīng)干細胞通過有絲分裂形成祖細胞(progenitorcell),后者遷移到嗅球,分化成新的中間神經(jīng)元,代替衰老或死亡的神經(jīng)細胞,使局部神經(jīng)環(huán)路的結構得到不斷更新,從而發(fā)揮正常的嗅覺及嗅覺記憶功能[1]。褪黑素是主要由松果體合成和分泌的一種神經(jīng)內分泌激素,已證實其不僅具有促進胚胎個體發(fā)育及體外培養(yǎng)神經(jīng)干細胞增殖的功能[2,3],而且還可提高學習記憶能力[4]。然而,迄今仍匱乏有關褪黑素對成年在體SVZ神經(jīng)發(fā)生及學習記憶影響的報道。為此,筆者最近進行了研
7、究并初步觀察到,松果體切除對成年大鼠SVZ神經(jīng)發(fā)生及學習記憶均產(chǎn)生類似的負性效應[5]。為進一步揭示褪黑素、神經(jīng)干細胞及學習記憶三者之間的內在聯(lián)系,本研究通過觀察褪黑素替代治療對上述指標的影響模式,為闡明學習記憶的神經(jīng)內分泌機制提供新的資料。1材料與方法1.1主要試劑小鼠抗人PA單克隆抗體為DAKO公司產(chǎn)品;生物素結合IgG、鏈霉菌抗生物素-過氧化酶(SP)及二氨基聯(lián)苯胺(DAB)染色試劑盒均為福州邁新公司產(chǎn)品;褪黑素為Sigma公司產(chǎn)品。1.
