1、C基因型HBV轉(zhuǎn)基因小鼠的制備作者:尤玉琴,黃黎珍,劉光澤,佟明華,李秀梅,胡蓮美,顧為望,孔祥平【摘要】 目的:建立C型HBV轉(zhuǎn)基因小鼠,為乙肝防治研究提供實驗動物模型.方法:采用受精卵顯微注射法,制備HBV轉(zhuǎn)基因小鼠,用PCR,ELISA,熒光定量PCR和免疫組化的方法研究HBV基因在轉(zhuǎn)基因小鼠體內(nèi)的整合、復制和表達情況.結(jié)果:注射受精卵2840枚,篩選注射后成活的受精卵共2256枚,注射成活率79.4%.注射后受精卵共移植假孕雌鼠85只,其中有42只懷孕,假孕雌鼠妊娠率49%,共產(chǎn)下F0代小鼠168只,PCR檢測共有14只小鼠陽性,其中ELISA檢測血清HBsAg陽性5只,HBe
2、Ag均陰性.熒光定量PCR血清HBVDNA顯示,2只小鼠的拷貝數(shù)分別為3.12×105copies/L,1.98×105copies/L,經(jīng)傳代產(chǎn)下F1代小鼠61只,PCR檢測HBVDNA陽性5只,ELISA檢測HBsAg陽性4只,其中肝臟HBsAg免疫組化陽性2只,HBcAg均陰性,且肝組織表達HBsAg為胞漿型.結(jié)論:構(gòu)建的C型HBV基因不但可以在轉(zhuǎn)基因小鼠體內(nèi)進行復制表達,而且可以遺傳給下一代小鼠.【關(guān)鍵詞】肝炎病毒乙型小鼠轉(zhuǎn)基因肝炎表面抗原乙型肝炎核心抗原乙型血清DNA11 【Abstract】AIM:TogenerateHBV(C)genometransgenicmiceto

7、eywords】hepatitisBvirusmicetransgenic,hepatitisBsurfaceantigens;hepatitisBcoreantigens;serum;DNA 0引言 根據(jù)HBV基因序列的差異>8%,可將HBV毒株分為不同的基因型,目前已確定有A-H8個基因型.不同基因型呈一定的地理區(qū)域性分布,并且HBV基因型與其流行特征、致病性、疾病治療及預后等方面存在十分緊密的關(guān)系[1-5].我國HBV的主
