1、BMP異位誘導(dǎo)成骨的Micro-CT評(píng)價(jià)作者:李丹 劉民,胡蘊(yùn)玉,畢龍,段永紅,金靜,王軍,劉山金【摘要】借助Micro-CT評(píng)價(jià)單純牛骨形態(tài)發(fā)生蛋白(bovinebonemorphogeneticprotein,bBMP)異位誘導(dǎo)成骨的長(zhǎng)期三維影像學(xué)及骨質(zhì)變化。(20±2)g昆明小鼠21只,麻醉后于雙側(cè)股部肌肉中植入bBMP各2mg,分別于1、2、4、6、8、10、12周各處死3只,切取誘導(dǎo)分化組織,5%戊二醛固定,行Micro-CT掃描和三維重建,運(yùn)用ABA專用骨骼分析軟件測(cè)定組織礦含量(tissuem
2、ineralcontent,TMC),組織骨密度(tissuemineraldensity,TMD),骨體積分?jǐn)?shù)(bonevolumefraction,BVF),結(jié)構(gòu)模型指數(shù)(structuremodelindex,SMI),骨小梁厚度(trabecularthickness,Tb.Th),骨小梁數(shù)量(trabecularnumber,Tb.N)及皮質(zhì)骨骨密度(bonemineraldensity,BMD)等參數(shù),運(yùn)用SPSS10.0統(tǒng)計(jì)軟件進(jìn)行統(tǒng)計(jì)學(xué)分析。bBMP從植入2周開(kāi)始逐漸形成一橢圓形骨組織塊,2
3、~4周,異位生成骨呈疏松的新生骨,4周時(shí)組織礦含量達(dá)第一個(gè)峰值,骨小梁數(shù)量最多;隨著觀察時(shí)間的延長(zhǎng)(6~12周),異位誘導(dǎo)生成的橢圓形骨組織內(nèi)部骨小梁逐漸吸收,數(shù)量減少,12周時(shí)骨小梁數(shù)量最少;而外層骨組織逐漸塑形成為皮質(zhì)骨,12周時(shí)骨礦含量值、骨小梁厚度、組織骨密度和皮質(zhì)骨骨密度均達(dá)最大值。說(shuō)明bBMP具有強(qiáng)大的異位骨誘導(dǎo)能力,血供不足時(shí),骨質(zhì)降解吸收;血供充足時(shí),骨質(zhì)逐漸成熟改建。12【關(guān)鍵詞】骨形態(tài)發(fā)生蛋白;異位成骨;骨誘導(dǎo);骨小梁;MicroCT Abstract:Toassessthe3-Dim
