1、嗅成鞘細(xì)胞移植對(duì)AD大鼠腦內(nèi)COX活性和線粒體超微結(jié)構(gòu)的影響作者:姚柏春孫天敏王金勇李勇王配軍李文春【摘要】目的探討嗅成鞘細(xì)胞(OECs)移植對(duì)阿爾茨海默病(AD)大鼠腦內(nèi)COX活性和線粒體超微結(jié)構(gòu)的影響。方法SD大鼠雙側(cè)海馬注射Aβ1~40,建立AD大鼠模型。實(shí)驗(yàn)動(dòng)物分為四組:正常對(duì)照組、AD模型組、OECs移植組、人工腦脊液(MCSF)注射組,每組12只。體外原代培養(yǎng)OECs并將其移植至AD大鼠雙側(cè)海馬。運(yùn)用行為學(xué)測(cè)試、組織化學(xué)、原位雜交結(jié)合圖像分析以及電鏡等技術(shù)和工具,觀察、比較各組大鼠學(xué)習(xí)記憶能力、海馬CA1區(qū)線粒
2、體細(xì)胞色素氧化酶(COX)活性、細(xì)胞色素氧化酶Ⅱ型亞基(COⅡ)mRNA表達(dá),以及CA1區(qū)神經(jīng)元線粒體超微結(jié)構(gòu)等指標(biāo)的變化。結(jié)果與正常對(duì)照組比較,AD模型組大鼠空間學(xué)習(xí)記憶能力、海馬CA1區(qū)線粒體COX活性及COⅡmRNA表達(dá)明顯降低(P<0.05),線粒體數(shù)量減少,結(jié)構(gòu)不清、腫脹、空泡樣變、嵴斷裂;OECs移植明顯上調(diào)上述指標(biāo)(P<0.05),并對(duì)線粒體超微結(jié)構(gòu)有明顯的保護(hù)作用。結(jié)論OECs移植通過改善AD大鼠學(xué)習(xí)記憶能力、提高海馬COX活性和COⅡmRNA表達(dá)以及保護(hù)線粒體等作用,對(duì)大鼠AD具有明顯的治療效
