2、,半定量逆轉(zhuǎn)錄聚合酶鏈反應(yīng)技術(shù)檢測(cè)功能相關(guān)的Tg基因和雙鏈復(fù)合蛋白8(Pax-8)基因的表達(dá).結(jié)果表明,10-12、10-10mol·L-1暴露組Tg分泌量較對(duì)照組顯著減少(p<0.05),而10-8mol·L-1BDE-47組Tg分泌量與對(duì)照組無顯著性差異(p>0.05).甲狀腺功能相關(guān)Tg基因和Pax-8基因的表達(dá)量隨BDE-47濃度的增加而顯著降低(p<0.05),呈明顯的劑量-效應(yīng)關(guān)系.以上結(jié)果提示,BDE-47對(duì)離體培養(yǎng)的原代人甲狀腺細(xì)胞的功能具有抑制作用,甲狀腺功能相關(guān)Tg基因和Pax-8基因的表達(dá)下調(diào)可能
