1、第四軍醫(yī)大學(xué)碩士學(xué)位論文學(xué)染色及Westernblotting方法檢測凋亡相關(guān)蛋白(CytoC、Smac)表達(dá)。結(jié)果:Rhodamine123染色顯示,800μmol/LPQ處理24h后,細(xì)胞的熒光強度與對照相比顯著降低(P<0.001)。而EGCG可劑量依賴性的減輕MMP的下降。與PQ組(26.2±2.8%)相比,經(jīng)1、5、10μmol/LEGCG預(yù)處理后,細(xì)胞熒光強度分別增至41.3±3.6%(P<0.01),56.7±5.4%(P<0.001),82.9±4.2%(P<0.001);PQ組與正常對照組相比,Caspase-
2、3活性顯著增強(P<0.001),而這種激活可被1、5、10μmol/LEGCG預(yù)處理所抑制。與PQ組(216.5±9.6)相比,經(jīng)EGCG預(yù)處理后,Caspase-3活性分別降至179.9±7.6(P<0.001)、123.2±8.1(P<0.001)、68.5±6.1(P<0.001);免疫細(xì)胞化學(xué)染色顯示正常組細(xì)胞形態(tài)良好,CytoC、Smac染色呈淡棕黃色,均勻分布于胞質(zhì)及突起內(nèi)。PQ處理后部分細(xì)胞變圓,皺縮,CytoC、Smac在細(xì)胞內(nèi)表達(dá)增強,染色呈深棕黃色。相對于PQ組,EGCG預(yù)處理組的CytoC、Smac免疫細(xì)
3、胞化學(xué)染色明顯減弱,細(xì)胞形態(tài)也有改善。Westernblotting檢測Smac蛋白表達(dá)結(jié)果表明PQ處理后胞漿內(nèi)Smac表達(dá)增加,EGCG預(yù)處理可以抑制這種改變。結(jié)論:EGCG對PQ誘導(dǎo)的PC12細(xì)胞凋亡具有保護(hù)作用,機制可能與其維持細(xì)胞MMP、抑制Caspase-3激活、調(diào)節(jié)凋亡相關(guān)蛋白CytoC及Smac在胞漿中的表達(dá)有關(guān)。關(guān)鍵詞:EGCG;百草枯;帕金森??;PC12細(xì)胞;細(xì)胞凋亡;神經(jīng)保護(hù)6第四軍醫(yī)大學(xué)碩士學(xué)位論文NeuroprotectiveeffectsofEGCGonparaquat-inducedapoptosis
