1、昆蟲(chóng)學(xué)報(bào)ActaEntomologicaSinica,March2007,50(3):241-247ISSN04546296煙粉虱對(duì)擬除蟲(chóng)菊酯殺蟲(chóng)劑的抗性機(jī)理何玉仙1,黃建2,楊秀娟1,翁啟勇1,*(1福建省農(nóng)業(yè)科學(xué)院植物保護(hù)研究所,福州350013;2福建農(nóng)林大學(xué)植物保護(hù)學(xué)院,福州350002)摘要:通過(guò)增效劑生物測(cè)定、生化分析以及鈉離子通道基因S46cDNA片段的RTPCR擴(kuò)增,探討了煙粉虱Bemisiatabaci(Gennadius)對(duì)擬除蟲(chóng)菊酯殺蟲(chóng)劑的抗性機(jī)理。結(jié)果表明:對(duì)于采自田間的6個(gè)煙粉虱抗性品系,磷酸三苯酯(TPP)和胡椒基丁
2、醚(PBO)對(duì)氯氰菊酯、溴氰菊酯、氯氟氰菊酯和甲氰菊酯均有顯著的增效作用,而DEM對(duì)4種擬除蟲(chóng)菊酯殺蟲(chóng)劑均無(wú)明顯的增效作用。煙粉虱抗性品系的NA羧酸酯酶和NA羧酸酯酶活性分別是敏感品系的216~265倍和122~141倍,抗性品系的谷胱甘肽S轉(zhuǎn)移酶活性與敏感品系沒(méi)有差異,表明羧酸酯酶和多功能氧化酶在煙粉虱對(duì)擬除蟲(chóng)菊酯類殺蟲(chóng)劑的抗性中具有重要的作用,而谷胱甘肽S轉(zhuǎn)移酶與抗性無(wú)關(guān)。通過(guò)RTPCR克隆了6個(gè)煙粉虱田間抗性品系的鈉離子通道結(jié)構(gòu)域S46cDNA片段的序列(420bp),發(fā)現(xiàn)與敏感品系相比,有2個(gè)位點(diǎn)發(fā)生突變,分別為L(zhǎng)925I突變和I917
