1、編號:200809111刊用形式:論著構(gòu)建人骨保護素在乳腺癌細(xì)胞中的表達(dá)張帆1,周艷1,唐鵬1,姜軍1(1第三軍醫(yī)大學(xué)西南醫(yī)院乳腺中心,重慶400038)摘要:目的建立過表達(dá)人骨保護素(OPG)的MDA-MB-231乳腺癌細(xì)胞克隆。方法 從pOTB7-OPG上獲得OPG基因片段并用PCR方法擴增,將其連接于真核表達(dá)質(zhì)粒pIRES2-EGFP,酶切鑒定及測序后,轉(zhuǎn)染MDA-MB-231細(xì)胞,以G418加壓篩選,對轉(zhuǎn)染細(xì)胞行單克隆化,穩(wěn)定轉(zhuǎn)染細(xì)胞行RT-PCR和Westernblot檢測,確定其OPGmRNA和蛋白表達(dá)情況,MTT法檢測過表達(dá)
2、OPG對MDA-MB-231細(xì)胞生長速率的影響。結(jié)果成功構(gòu)建了pIRES2-EGFP-OPG重組質(zhì)粒;穩(wěn)定轉(zhuǎn)染細(xì)胞的OPGmRNA和蛋白表達(dá)均較對照升高;過表達(dá)OPG對MDA-MB-231細(xì)胞生長速率無明顯影響。結(jié)論本研究成功建立了過表達(dá)OPG的MDA-MB-231細(xì)胞克隆,為進一步深入探討腫瘤細(xì)胞自身OPG表達(dá)在乳腺癌骨轉(zhuǎn)移發(fā)生發(fā)展中的作用提供實驗基礎(chǔ)。關(guān)鍵詞:乳腺癌;骨保護素;過表達(dá);MDA-MB-231細(xì)胞;pIRES2-EGFP中圖法分類號:文獻標(biāo)識碼:AHumanosteoprotegerinconstructedandexpr
