1、提要目的:探討納米雄黃對(duì)人肺腺癌A549細(xì)胞株血管內(nèi)皮生長(zhǎng)因子(VEGF)、缺氧誘導(dǎo)因子(HIF-1)表達(dá)的影響及其抗腫瘤血管生成的機(jī)理。方法:采用細(xì)胞培養(yǎng)方法,運(yùn)用免疫組化法、RT-PCR技術(shù)觀察納米雄黃對(duì)人肺癌A549細(xì)胞VEGF、HIF-1的表達(dá)的影響,進(jìn)而探討納米雄黃抗腫瘤血管生成的作用機(jī)理。結(jié)果:實(shí)驗(yàn)研究證實(shí),納米雄黃可以抑制人肺癌A549細(xì)胞增殖,隨藥物濃度的增高作用增強(qiáng),且與順鉑聯(lián)合應(yīng)用有協(xié)同作用,初步揭示納米雄黃抗腫瘤血管生成的機(jī)理可能與下調(diào)VEGF、HIF-1的表達(dá)有關(guān)。結(jié)論:納米雄黃可以通過抑制腫瘤細(xì)胞血管生成等途徑實(shí)現(xiàn)抗腫瘤作用,與順鉑聯(lián)合可產(chǎn)生協(xié)同作
