1、富硒金針菇菌種的選育摘要目的:食用菌以其獨(dú)特的營(yíng)養(yǎng)價(jià)值、藥理作用和簡(jiǎn)易高效的生產(chǎn)工藝等優(yōu)勢(shì)備受青睞。金針菇(Flammnlinavelutipes)作為食用菌中的一朵奇葩,更有美味可口、營(yíng)養(yǎng)豐富、抗癌治潰瘍等作用。目前對(duì)金針菇的營(yíng)養(yǎng)成分報(bào)道較多,本文對(duì)金針菇菌絲體及其紫外誘變體的最佳富硒能力的培養(yǎng)基硒含量、菌絲生長(zhǎng)特性、平板富硒培養(yǎng)與液體富硒培養(yǎng)、菌絲體硒含量等做了探討。方法:本實(shí)驗(yàn)配置馬鈴薯綜合培養(yǎng)基、紫外菌絲體誘變、富硒CM培養(yǎng)基培養(yǎng)金針菇201和白色金針菇;針對(duì)固體平板培養(yǎng),采用十字交叉法測(cè)量菌落半徑,使用乳酸石炭酸棉藍(lán)染色液將菌絲染色,測(cè)其菌絲直徑;針對(duì)液體發(fā)酵,使用游標(biāo)卡尺測(cè)量菌絲
2、球直徑,收集菌絲球于稱量瓶,在電熱恒溫干燥箱中烘干至恒重,用電子分析天平稱其干重;使用紫外分光光度法測(cè)定菌絲含硒量;對(duì)實(shí)驗(yàn)數(shù)據(jù)的處理采用EXCEL制圖、SPSS統(tǒng)計(jì)分析處理。結(jié)果:白色金針菇、金針菇201及其誘變菌種均能在含硒量為20~35μg/L的培養(yǎng)基上生長(zhǎng)速度達(dá)到最快,富硒能力最強(qiáng)的狀態(tài)。結(jié)論:相對(duì)于固體平板培養(yǎng),液體發(fā)酵培養(yǎng)菌絲體的生長(zhǎng)速度及富硒能力達(dá)到新的高度。關(guān)鍵詞:金針菇;硒;紫外誘變;固化富硒培養(yǎng);液體搖床培養(yǎng)IIITheBreedingofSelenium-accumulatingStrainofFlammulinavelutipessAbstractSubject:Edi
