6、····44個(gè)人簡(jiǎn)歷·············································································45中文摘要EC-SOD在大鼠肝臟缺血再灌注損傷中的表達(dá)變化摘要臨床上在進(jìn)行肝臟手術(shù)如肝臟部分切除、肝臟移植以及處理嚴(yán)重肝臟外傷時(shí)往往需要暫時(shí)性阻斷肝臟血流一段時(shí)間后再重新恢復(fù)其血液供應(yīng)。在這個(gè)過(guò)程中就造成了肝臟的缺血再灌注損傷(hepaticischemiareperfusioninjury,HIRI)。HIRI是肝臟外科手術(shù)過(guò)程中一種常見(jiàn)的病理生理現(xiàn)象。肝臟的缺血再灌注可引起嚴(yán)重的肝細(xì)胞損傷

7、和肝功能障礙,這也是導(dǎo)致術(shù)后肝功能衰竭和病人預(yù)后較差的重要原因之一。研究已證實(shí),導(dǎo)致這種損傷的主要機(jī)制之一為氧化應(yīng)激反應(yīng)。缺血再灌注時(shí)機(jī)體會(huì)產(chǎn)生大量活性氧自由基(reac-tiveoxygenspecies,ROS)。ROS包括超氧陰離子-(O2)、羥自由基(OH)和過(guò)氧化氫(H2O2)等。ROS的化學(xué)性質(zhì)非?;顫?,可以破壞細(xì)胞內(nèi)的蛋白質(zhì)、DNA和脂類等生物大分子,引起細(xì)胞死亡。超氧化物歧化酶(SuperoxideDismutases,SOD)是機(jī)體內(nèi)一種重要--的抗氧化酶,負(fù)責(zé)清除O2,它能將O2轉(zhuǎn)化成H2O2和氧,在機(jī)體抗氧化應(yīng)激反應(yīng)中具有重要作用。在哺乳動(dòng)
