6、·············46個(gè)人簡(jiǎn)歷·············································································48中文摘要YAP與P-YAP在肝細(xì)胞癌中的表達(dá)與其臨床生物學(xué)行為關(guān)系的研究摘要目的:肝細(xì)胞癌(HCC,hepatocellularcarcinoma)是肝臟原發(fā)腫瘤中最常見的類型,也是我國(guó)乃至全世界.最常見的惡性腫瘤疾病之一。通常HCC被發(fā)現(xiàn)時(shí)患者已進(jìn)入晚期,常規(guī)化療對(duì)其作用不明顯,致使生存率較其它癌癥顯著降低。因此,加強(qiáng)對(duì)HCC的發(fā)病機(jī)制研究,尋找更多關(guān)于肝癌基因及抑癌基因的表達(dá)并明確其與

7、HCC的關(guān)系已成為當(dāng)前研究的重要方向。Hippo通路在促進(jìn)細(xì)胞凋亡,限制細(xì)胞增殖,調(diào)控器官分化方面起著重要的作用。YAP(Yes-associatedprotein)即Yes相關(guān)蛋白,作為該通路下游的效應(yīng)因子,多以磷酸化形式(P-YAP)參與細(xì)胞信號(hào)轉(zhuǎn)導(dǎo)以調(diào)控細(xì)胞凋亡與增殖的平衡。在病理狀態(tài)下,被阻斷或失活的Hippo通路對(duì)YAP磷酸化作用失效致YAP自身活性增強(qiáng)使細(xì)胞增生和凋亡間平衡失調(diào)、組織器官過度生長(zhǎng)及腫瘤發(fā)生。國(guó)內(nèi)外相關(guān)研究表明,YAP作為新近發(fā)現(xiàn)的癌基因,參
