1、黃芪多糖對(duì)小鼠樹(shù)突狀細(xì)胞前體的體內(nèi)誘導(dǎo)研究作者:胡微微,臧國(guó)慶,余永勝【摘要】目的探討黃芪多糖(astragaluspolysaccharide,APS)在體內(nèi)是否具有誘導(dǎo)小鼠髓源性樹(shù)突狀細(xì)胞(dendriticcell,DC)分化和成熟作用,以闡釋APS藥理作用機(jī)制。方法分別設(shè)對(duì)照組及APS100、200mg/kg劑量組,腹腔注射,體內(nèi)誘導(dǎo)小鼠髓源性DC前體和未成熟DC,稱量脾重,ELISA檢測(cè)血清中GM-CSF表達(dá)量,倒置顯微鏡觀察細(xì)胞生長(zhǎng)狀態(tài),電鏡檢測(cè)細(xì)胞形態(tài),流式細(xì)胞儀檢測(cè)CD11c、MHC-Ⅱ類分子
2、及協(xié)同刺激分子CD80、CD86表達(dá)水平。結(jié)果APS注射7d后,APS100、200mg/kg組小鼠脾臟重量均較對(duì)照組有明顯增加(P<0.01),骨髓細(xì)胞體外分離并經(jīng)rmGM-CSF、rmIL-4誘導(dǎo)培養(yǎng)后,流式細(xì)胞檢測(cè)結(jié)果顯示:APS100、200mg/kg組CD11c、MHC-Ⅱ類分子表達(dá)水平高于對(duì)照組(P<0.05),但CD80、CD86表達(dá)量較對(duì)照組無(wú)明顯差異;血清中GM-CSF表達(dá)量,結(jié)果與對(duì)照組無(wú)明顯差異。結(jié)論APS能誘導(dǎo)小鼠髓源性DC前體分化為DC,并可能有促進(jìn)DC成熟的作用,但此作

8、agaluspolysaccharide;mice樹(shù)突狀細(xì)胞(DC)是目前發(fā)現(xiàn)體內(nèi)最強(qiáng)的抗原提呈細(xì)胞,能激活初始T細(xì)胞,在誘導(dǎo)特異性免疫反應(yīng)過(guò)程中起著重要作用[1]。腫瘤、慢性肝炎、自身免疫性疾病及移植排斥反應(yīng)等疾病的發(fā)生、發(fā)展與患者免疫功能密切相關(guān),有研究證明,這些患者體內(nèi)DC發(fā)生了不同程度的數(shù)量和功能變化,因此,通過(guò)改變DC數(shù)量及成熟狀態(tài),糾正機(jī)體免疫紊亂,進(jìn)而達(dá)到治療和控制疾病的目的。黃芪多糖
