1、脂肪來源的干細(xì)胞移植對(duì)大鼠腦缺血后微血管生成及bFGF和VEGF表達(dá)的影響x作者:王杰華,劉楠,杜厚偉,翁金森,陳榮華,肖迎春,張逸仙【摘要】目的:探討脂肪來源的干細(xì)胞(ADSC)移植對(duì)大鼠腦缺血后微血管生成的可能機(jī)制。方法:72只清潔級(jí)成年雄性SpragueDarley大鼠,隨機(jī)分為假手術(shù)組(Sham組)、局灶性腦缺血組(MCAO組)、溶劑對(duì)照組(vehicle組)和ADSC治療組(ADSC組),每組18只,采用改良ZeaLonga線栓法制作大腦中動(dòng)脈栓塞(MCAO)模型,ADSC移植前用DAPI標(biāo)記,ADSC組于造模成功1d后經(jīng)側(cè)腦室注射入30μL的經(jīng)DAPI標(biāo)記的ADS
2、C細(xì)胞懸液,內(nèi)含1×106細(xì)胞,vehicle組則注射同等劑量的PBS,術(shù)后4d、7d和14d分批處死實(shí)驗(yàn)動(dòng)物,并斷頭取腦,采用免疫組織化學(xué)法和半定量RTPCR法檢測(cè)缺血區(qū)腦組織的新生血管及bFGF和VEGF表達(dá)的動(dòng)態(tài)變化。結(jié)果:大鼠腦缺血后缺血區(qū)即可見大量的微血管生成,2周達(dá)高峰。ADSC組較MCAO組和vehicle組缺血區(qū)微血管密度明顯增高(P<0.01);ADSC組術(shù)后4d、7d和14d腦組織中bFGF和VEGF的表達(dá)水平較MCAO組和vehicle組明顯增高。結(jié)論:ADSC移植促進(jìn)腦缺血大鼠缺血區(qū)微血管的生成,其機(jī)制可能與促進(jìn)bFGF和VEGF的表達(dá)有關(guān)【關(guān)鍵詞
