1、萬(wàn)方數(shù)據(jù)史堡超絲匡堂參圭一2--010生墨旦箜芝鲞一笪8勞JChinJNeuromed,August2010,V01.9,No.8CNQX對(duì)乙醇、乙醛調(diào)控大腦皮質(zhì)神經(jīng)元神經(jīng)甾體水平的影響張翠欣侯艷寧劉紅衛(wèi)張彤【摘要】目的研究乙醇、乙醛對(duì)原代培養(yǎng)大鼠大腦皮質(zhì)神經(jīng)元神經(jīng)甾體水平的影響以及與AMPA受體調(diào)控的相關(guān)性。方法應(yīng)用AMPA受體拮抗劑CNQX阻斷AMPA受體.用50mlnol/L乙醇或25mmol/L乙醛處理神經(jīng)元,收集細(xì)胞培養(yǎng)上清,采用高效液相色譜質(zhì)譜法分離測(cè)定游離型神經(jīng)甾體孕烯醇酮(PREG)、脫氫表雄酮(DHEA)、別孕烯醇酮(AP)及結(jié)合型神經(jīng)甾體脫
2、氫表雄酮硫酸酯(DHEAS)、孕烯醇酮硫酸酯(PREGS)的含量。觀察CNQX對(duì)乙醇、乙醛調(diào)控原代培養(yǎng)神經(jīng)元神經(jīng)甾體水平的影響。結(jié)果乙醇可明顯升高神經(jīng)元PREG、AP和PREGS的水平,與PBS對(duì)照組比較差異有統(tǒng)計(jì)學(xué)意義(風(fēng)0.05);乙醛可明顯升高神經(jīng)元PREG、DHEA、PREGS和DHEAS的水平,與PBS對(duì)照組比較差異有統(tǒng)計(jì)學(xué)意義職O.05)。CNQX反轉(zhuǎn)了乙醇升高PREG、PREGS水平的作用以及乙醛升高PREG、DHEAS水平的作用。卻使AP水平明娃升高。組間比較差異有統(tǒng)計(jì)學(xué)意義(氏0.05)。結(jié)論乙醇、乙醛對(duì)大腦皮質(zhì)神經(jīng)元神經(jīng)甾體PREG、DHE
