1、厄多司坦預(yù)處理對大鼠心肌缺血-再灌注損傷及心肌細胞凋亡的影響王洪波,汪斌,向小勇(400016重慶,重慶醫(yī)科大學(xué)附屬第一醫(yī)院胸心外科)[摘要]目的探討厄多司坦預(yù)處理對大鼠心肌缺血-再灌注損傷(myocardialischemia-reperfusioninjury,MIRI)的保護作用及對心肌細胞凋亡的影響。方法雄性SD大鼠30只,完全隨機設(shè)計法分成偽手術(shù)組、缺血-再灌注組及厄多司坦預(yù)處理組,每組10只。采用結(jié)扎左冠狀動脈前降支30min、再灌注120min的方法建立大鼠心肌缺血-再灌注損傷模型。再灌注末,測定血清丙二醛(MDA)含量,超氧化物歧化酶
2、(SOD)、過氧化氫酶(CAT)、谷胱甘肽過氧化酶(GSH-Px)、肌酸激酶(CK)、乳酸脫氫酶(LDH)的活性;心肌MDA含量,SOD、CAT、GSH-Px的活性;TUNEL染色檢測心肌細胞凋亡。結(jié)果厄多司坦預(yù)處理減少再灌注末血清及心肌MDA含量(P<0.05),降低血清CK、LDH活性(P<0.05,P<0.01),提高血清及心肌SOD、CAT及GSH-Px活性(P<0.05,P<0.01),降低心肌細胞凋亡率(P<0.05)。結(jié)論厄多司坦預(yù)處理可減輕在體心臟缺血-再灌注模型大鼠的氧化應(yīng)激損傷,并可減輕氧自由基誘導(dǎo)的心肌細胞凋亡。[關(guān)鍵詞]厄多司
