2、生,為刺槐生產(chǎn)及育種提供參考依據(jù)?!痉椒ā恳源袒被ǚ蹫樵嚥?,使用培養(yǎng)基萌發(fā)試驗(yàn)法測(cè)定其生活力,并研究不同濃度ca”、不同貯藏條件及時(shí)間對(duì)刺槐花粉萌發(fā)的影響?!窘Y(jié)果】刺槐花粉在瓊脂8.0g/L
3、+蔗糖20.0mg/L+硼酸2.0mg/L的培養(yǎng)基中萌發(fā)率高達(dá)30.25%;適當(dāng)濃度的ca2_能促進(jìn)花粉萌發(fā)和花粉管伸長(zhǎng),當(dāng)caz榷度為1.6x10。mg/L時(shí),花粉萌發(fā)率高達(dá)40.25%,花粉管伸長(zhǎng)最快;適宜刺槐花粉貯藏的條件依次為名o℃、4℃、一23℃和常溫;花粉在名o℃貯藏32d后,萌發(fā)率仍達(dá)15.30%?!窘Y(jié)論】適合刺槐花粉萌發(fā)的培養(yǎng)基為:瓊脂8.0g/L+蔗糖20.0mg/L+硼酸2.0mg/L;培養(yǎng)基中添加低濃度外源鈣能促進(jìn)刺槐花粉萌發(fā)和花粉管生長(zhǎng);于超低溫真空冷凍條件下貯藏的刺槐花粉保持活力時(shí)間更長(zhǎng)。關(guān)鍵詞:刺槐;花粉活力;貯藏條件;培養(yǎng)基萌發(fā)法
