2、35(4):1059鄄1067.基于ISSR分子標(biāo)記的褐馬雞親緣關(guān)系分析123,*武玉珍,馮睿芝,張搖峰1晉中學(xué)院生物科學(xué)與技術(shù)學(xué)院,晉中搖0306192復(fù)旦大學(xué)生物醫(yī)學(xué)研究院,上海搖2000323山西大學(xué)生命科學(xué)院,太原搖030006摘要:褐馬雞(Crossoptilonmantchuricum)是中國(guó)特有瀕危鳥(niǎo)類(lèi),國(guó)家一級(jí)保護(hù)動(dòng)物。為了保護(hù)褐馬雞種質(zhì)資源,保障馴養(yǎng)繁育種群和再引入種群的遺傳基因結(jié)構(gòu)優(yōu)化,采用ISSR分子標(biāo)記技術(shù),對(duì)山西龐泉溝國(guó)家自然保護(hù)區(qū)和太原動(dòng)物園兩個(gè)種群35個(gè)褐馬雞進(jìn)行了親緣關(guān)系分析。從20條ISSR引物中篩選出10條擴(kuò)增條帶清晰穩(wěn)定、重復(fù)性好的引物
3、,共擴(kuò)增出65條DNA條帶,其中77%呈多態(tài)性。兩個(gè)種群各個(gè)體之間的Nei憶s無(wú)偏差遺傳距離和遺傳相似度分析表明:個(gè)體間遺傳相似性的平均值分別為0.5061與0.7591,遺傳距離的平均值分別為0.4939與0.2409。用組平均法對(duì)35個(gè)褐馬雞個(gè)體進(jìn)行聚類(lèi)分析,結(jié)果顯示:大多數(shù)同一種群的個(gè)體首先聚在一起,顯示出生態(tài)地理?xiàng)l件相似的種群親緣關(guān)系較近。關(guān)鍵詞:褐馬雞;ISSR;遺傳距離;遺傳相似度;聚類(lèi)分析;親緣關(guān)系A(chǔ)nalysisongeneticrelationshipofBrown鄄earedpheasant(Crossoptilonmantchuricum)basedo
