2、，并觀察rhlL-24對(duì)卵巢癌SKOV3和SKOV3／DDP細(xì)胞凋亡和細(xì)胞周期的影響。方法用rhlL-24、DDP及rhlL-24聯(lián)合DDP作用于SKOV3、SKOV3／DDP細(xì)胞，以CCK-8法檢測(cè)細(xì)胞增殖、用流式細(xì)胞儀檢測(cè)細(xì)胞凋亡及周期的變化。結(jié)果rhlL一24對(duì)體外培養(yǎng)的卵巢癌SKOV3和SKOV3／DDP細(xì)胞的生長(zhǎng)有明顯抑制作用，rhlL-24聯(lián)合DDP對(duì)SKOV3／DDP細(xì)胞株抑制率為30．7％，與單用DDP組抑制率6．5％相比有顯著性差異，流式細(xì)胞儀檢測(cè)rhlL-24、DDP和rhlL-24聯(lián)合DDP各
3、組凋亡率分別為14．95％、12．99％和16．32％，與陰性對(duì)照組凋亡率1．32％比較具有顯著性差異；細(xì)胞周期檢測(cè)中，SKOV3細(xì)胞周期變化顯示rhlL-24組G2、s期增多，聯(lián)合用藥組出現(xiàn)s期增多；SKOV3／DDP細(xì)胞中，rhlL-24組處理出現(xiàn)G2期增多，聯(lián)合用藥組出現(xiàn)G1期增多。結(jié)論rhlL-24對(duì)卵巢癌細(xì)胞DDP敏感和耐藥株都具有抑制作用，聯(lián)合用藥可增強(qiáng)DDP的效應(yīng)；rhlL-24對(duì)卵巢癌細(xì)胞的抑制作用通過誘導(dǎo)細(xì)胞凋亡實(shí)現(xiàn)，rML-24處理細(xì)胞后周期變化出現(xiàn)G2期阻滯，聯(lián)合用藥組出現(xiàn)G1期阻滯。[關(guān)鍵詞
