1、營養(yǎng)與僳1j辜[7o1.35,JVo.13,2014李子提取物誘導(dǎo)人肝癌HepG2細(xì)胞的凋亡及其作用機(jī)制楊成流。劉金娟。蔣繼宏(江蘇師范大學(xué)江蘇省藥用植物生物技術(shù)重點(diǎn)實(shí)驗(yàn)室,江蘇徐州221116)摘要:探討李子提取物對人肝癌細(xì)胞HepG2增殖和凋亡的影響及其作用機(jī)制。采用AlamarBlue法檢測李子提取物對HepG2細(xì)胞增殖的抑制作用;倒置顯微鏡、Hoechst33258熒光染色、流式細(xì)胞術(shù)檢測李子提取物對HepG2細(xì)胞凋亡的影響;Westernblot分析李子提取物誘導(dǎo)HepG2細(xì)胞凋亡的作用機(jī)制。結(jié)果表明,5%和10%的李子提取物作用HepG2細(xì)胞48h后,明顯抑制He
2、pG2細(xì)胞的增殖;細(xì)胞出現(xiàn)明顯的凋亡特征,流式細(xì)胞術(shù)進(jìn)一步證實(shí)李子提取物能夠誘導(dǎo)HepG2細(xì)胞的凋亡;處理組HepG2細(xì)胞中Bcl一2和Survivin表達(dá)量降低,p53,Bax,Bid,Caspase一9和Caspase一3表達(dá)量增加并且Caspase一3酶活性顯著提高。李子提取物通過內(nèi)源性凋亡信號通路誘導(dǎo)HepG2細(xì)胞的凋亡發(fā)揮其抗腫瘤活性。關(guān)鍵詞:李子提取物,肝癌細(xì)胞HepG2,細(xì)胞凋亡,分子機(jī)制TheapoptosisofHepG2cellsinducedbyplumextractanditsmechanismYANGCheng—liu,LIUJin—juan,JIA

7、0306(2014)13—0359—04doi:10.13386/j.issnl002—0306.2014.13.069肝癌是一種在世界范圍內(nèi)最常見且死亡率很高我國栽培廣泛,具有豐富的營養(yǎng)和藥用價(jià)值。目前的惡性腫瘤。目前,最佳的治療方法認(rèn)為是手術(shù)切對李子的研究大多集中在其揮發(fā)性物質(zhì)方面。有除,但手術(shù)方法復(fù)發(fā)率高且適宜的人群較少?。因研究資料表明,李子果肉中含有單寧、酚類、多糖等此,尋找更有效的治療和預(yù)防這種惡性腫瘤的藥物物質(zhì),具有抗氧化、降血壓、降血糖、治療尿結(jié)石,促就很有必要。最近,利用飲食中常見的物
