1、廣東藥學(xué)院學(xué)報JournalofGuangdongPharmaceuticalUniversityFeb.2014.30(1)聚乳酸聚乙醇酸微球冷凍切片方法的研究黎吶,麥海燕,羅宇燕,鐘晨,張永明(1.中山大學(xué)藥學(xué)院,廣東廣州510006;2.中山大學(xué)附屬第三醫(yī)院,廣東廣州510630)摘要:目的研究聚乳酸聚乙醇酸(PLGA)微球切片的制備方法,以便使用掃描電鏡獲得孔洞結(jié)構(gòu)完整清晰的微球截面圖片,為后期使用圖像處理軟件計算微球的孔隙率提供基礎(chǔ)。方法使用冷凍包埋劑包埋載有牛血清白蛋白(BSA)的PLGA微球,冷凍切片獲得微球截面,并用掃描電子顯微鏡觀
2、察切片形態(tài),分別考察包埋劑的種類、浸泡時間、液氮冷凍等對切片質(zhì)量的影響。結(jié)果優(yōu)化方法為:將微球浸泡在自制的質(zhì)量濃度10%明膠+5%甘油混合冷凍包埋劑中,于37cc浸泡3h,然后轉(zhuǎn)移至質(zhì)量濃度20%明膠+5%甘油混合冷凍包埋劑中,于37℃浸泡1h,再轉(zhuǎn)移至一20℃的冰箱中使包埋劑凝固;切片前將包埋塊浸入液氮中急速冷凍5min。結(jié)論使用優(yōu)化方法所得的PLGA微球切片,截面孔}同結(jié)構(gòu)完整清晰,且操作方法簡單、快速和有效。關(guān)鍵詞:聚乳酸聚乙醇酸:微球;微觀結(jié)構(gòu);冷凍切片技術(shù)中圖分類號:R944文獻標(biāo)志碼:Adoi:10.3969/j.issn.1006—8
