1、·540·中草焉ChineseTraditionalandHerbalDrugs第43卷第3期2012年3月羅布麻葉總黃酮對高脂高鹽大鼠高血壓的影響及其分子機制李芝1,2王超云,張樹平,王軍蓮,張小敏,賀倩倩1.濱州醫(yī)學院藥學院,山東煙臺2640032.浙江大學醫(yī)學院,浙江杭州310031摘要:目的觀察羅布麻葉總黃酮的降壓作用,探討其作用機制。方法50只雄性SD大鼠隨機分為對照組,模型組,羅布麻葉總黃酮高、低劑量(32、8mg/kg)組和羅布麻降壓片(0.4g/kg)陽性對照組。除對照組外,其他各組在喂飼高脂高鹽飼料誘導大鼠高血壓模型的同時,每天ig給予不同劑量的藥物,并運
2、用無創(chuàng)血壓測量儀監(jiān)測大鼠血壓的變化。造模6周后,利用免疫組化法檢測血管組織轉化生長因子.131(TGF。131)蛋白表達,Westernblotting法檢測C一反應蛋白(CRP)表達,分光光度法檢測血清中高密度脂蟹白.膽固醇(HDL.C)、低密度脂蛋白.膽尉醇(LDL—C)、總膽固醇(TC)、三酰甘油(TG)水平。結果高脂高鹽飼料能顯著升高大鼠血壓,改變血管組織中CRP、TGF.131的表達。與模型組比較,羅布麻葉總黃酮在高劑量時能降低血壓,抑制血管組織中CRP、TGF.131的表達,提高血清中HDL-C的水平,降低LDL-C、TC、TG的水平。結論羅布麻葉總黃酮具有顯著
