3、要目的:通過對(duì)大鼠缺血缺氧性腦損傷模型注射不等含量的神經(jīng)生長因子,并和正常大鼠模型的不同部位神經(jīng)生長因子濃度和受體含量進(jìn)行對(duì)比,得出神經(jīng)生長因子和其受體在缺血缺氧性腦損傷中的作用機(jī)制,為臨床指導(dǎo)治療缺血缺氧性腦損傷提供依據(jù)。方法:1.大鼠模型資料:選取南方醫(yī)科大學(xué)動(dòng)物中心的16-18周大鼠若干只,雌雄不限,模型均隨機(jī)篩選,統(tǒng)計(jì)結(jié)果無統(tǒng)計(jì)學(xué)差異。2.方法:將這些隨機(jī)抽取的大鼠模型一共50只,按照是否手術(shù)和手術(shù)注射神經(jīng)生長因子的情況的不同分為5組,A組的大鼠模型正常喂養(yǎng),B組的大鼠模型做左頸動(dòng)脈結(jié)扎手術(shù),并且在持續(xù)喂養(yǎng)
4、時(shí)每日注射生理鹽水;C組的大鼠模型做左頸動(dòng)脈結(jié)扎手術(shù),并且在持續(xù)喂養(yǎng)時(shí)每日注射鼠神經(jīng)生長因子30ug;D組的大鼠模型做左頸動(dòng)脈結(jié)扎手術(shù),并且在持續(xù)喂養(yǎng)時(shí)每日注射鼠神經(jīng)生長因子60ug;E組的大鼠模型做左頸動(dòng)脈結(jié)扎手術(shù),并且在持續(xù)喂養(yǎng)時(shí)每日注射鼠神經(jīng)生長因子90ug,比較這五組大鼠模型在術(shù)后1天、術(shù)后一周、術(shù)后兩周、術(shù)后四周這幾個(gè)時(shí)間段頭部、腹部和尾部不同部位神經(jīng)生長因子濃度和TrkB受體含量的變化情況。3.數(shù)據(jù)分析:各組大鼠模型的基本資料情況經(jīng)正態(tài)性檢查,服從正態(tài)分布,且各組間無統(tǒng)計(jì)學(xué)差異。使用SAS6.12統(tǒng)計(jì)學(xué)
5、軟件按照2*2析因設(shè)計(jì)進(jìn)行統(tǒng)計(jì)學(xué)分析,分別通過對(duì)大鼠模型術(shù)后的統(tǒng)計(jì)指標(biāo)結(jié)果做出評(píng)估。數(shù)值型變量以均數(shù)±標(biāo)準(zhǔn)差表示。結(jié)果:五組大鼠模型在術(shù)后相同時(shí)間內(nèi)同組不同部位的神經(jīng)生長因子濃度和TrkB受體含量的統(tǒng)計(jì)學(xué)差異具有統(tǒng)計(jì)學(xué)意義(P<0.05);在術(shù)后相同時(shí)間內(nèi)不同組別同部位的神經(jīng)生長因子濃度和TrkB受體含量的統(tǒng)計(jì)學(xué)差異具有統(tǒng)計(jì)學(xué)意義(P<0.05);在術(shù)后不同時(shí)間內(nèi)同組別同部位的神經(jīng)生長因子濃度和TrkB受體含量的統(tǒng)計(jì)學(xué)差異具有統(tǒng)計(jì)學(xué)意義(P<0.05)。關(guān)鍵詞:神經(jīng)生長因子;TrkB受體;缺血缺氧性腦損傷;作用機(jī)制
