1、第43卷第3期西部林業(yè)科學(xué)Vol.43No.32014年6月JournalofWestChinaForestryScienceJun.20144個(gè)滇牡丹天然居群遺傳多樣性的ISSR分析*1222劉通,馮丹,陳少瑜,趙一鶴(1.西南林業(yè)大學(xué)環(huán)境科學(xué)與工程學(xué)院,云南昆明650224;2.云南省林業(yè)科學(xué)院,云南昆明650201)摘要:以云南省香格里拉縣4個(gè)滇牡丹天然居群為研究對(duì)象,采用ISSR分子標(biāo)記技術(shù),對(duì)98份材料進(jìn)行遺傳多樣性分析,用多態(tài)性高、穩(wěn)定性強(qiáng)的10條引物進(jìn)行擴(kuò)增,共獲得96條擴(kuò)增產(chǎn)物,其中多態(tài)
2、性條帶有82條,多態(tài)性百分比為85.42%;滇牡丹總體基因多樣性為0.3438,Shannon指數(shù)為0.5009;通過(guò)Nei’s和聚類(lèi)分析,居群間的遺傳分化系數(shù)為0.7973,居群間的遺傳變異占總遺傳變異的79.73%,而居群內(nèi)的遺傳變異只有20.27%,表明滇牡丹居群間的遺傳分化較大,遺傳變異主要存在于居群間。因此,滇牡丹雖然為分布區(qū)狹窄的特有種,但是與一些典型稀有和瀕危的物種相比,遺傳多樣性并不低,滇牡丹的分布區(qū)域和種群數(shù)量呈現(xiàn)狹窄化、縮小化的趨勢(shì),主要原因是受人為過(guò)度采挖,生境受到破壞所致。關(guān)鍵詞
