2、包裹質(zhì)粒pDHt-gpdA-GFP-bar,運用基因槍法轉(zhuǎn)化,經(jīng)草銨膦抗性篩選,最終在靶距離6cm或9cm,氦氣壓力66-17.58×10Pa或8.96×10Pa條件下,獲得2個遺傳穩(wěn)定的轉(zhuǎn)化子Gfp2與Gfp3,轉(zhuǎn)化效率為0.4cfu·μg。PCR鑒定與Southern雜交分析顯示,草銨膦抗性基因Bar已經(jīng)以單拷貝或者多拷貝方式整合到蛹蟲草轉(zhuǎn)化子的基因組中。蛹蟲草轉(zhuǎn)化子的菌絲在熒光顯微鏡下可以觀測到綠色熒光,表明載體攜帶的綠色熒光蛋白基因在蛹蟲草轉(zhuǎn)化子中得到表達。研究結果表明可以把基因槍法應用于真菌蛹蟲草,建立了一種簡便有效的遺傳轉(zhuǎn)化方法,而轉(zhuǎn)化效率需要
