1、蘭州大學(xué)磺士學(xué)位論文丙泊酚對(duì)大鼠肝臟缺血/再灌注損傷c-fos、c—jun及細(xì)胞凋亡的影響摘要目的C-fos、c-iun基因是一種快速的早期反應(yīng)基因(InlmediaIclyearlygene,lEG),在外界刺激一轉(zhuǎn)錄耦聯(lián)的信息傳遞過(guò)程中起著核內(nèi)第三信使的作用【11。丙泊酚對(duì)肝臟HIRI有保護(hù)作用,本實(shí)驗(yàn)通過(guò)研究丙泊酚對(duì)肝組織缺血再灌注損傷時(shí)C-los、C-jun蛋白的影響,進(jìn)一步闡明丙泊酚的作用機(jī)理,并將研究結(jié)果應(yīng)用到l臨床中,可更有效、更安全地完成麻醉、手術(shù)工作,合理用藥。方法選36只健康SD大鼠(雌雄不限)制作肝臟缺血/再灌注模型,分為3組:
2、肝臟缺血/再灌注組:肝缺血前30min經(jīng)微量泵持續(xù)輸注生理鹽水10ml·kg-1·F1至再灌注后lh;肝臟缺血,再灌注+丙泊酚組:肝缺血前經(jīng)微量泵持續(xù)輸注丙泊酚20m1.k91·h-1至再灌注后1h;假手術(shù)組:未予缺血處理,經(jīng)微量泵持續(xù)輸注生理鹽水lOm1.kg一·h.1共1.5h.各組大鼠取肝臟右葉標(biāo)本,免疫組化染色,顯微鏡下觀察C-fos、C0蛐的表達(dá),電鏡下觀察細(xì)胞凋亡情況。結(jié)果1、免疫組化結(jié)果顯示C-los、c.j蚰在缺血/再灌注+丙泊酚組的表達(dá)較單純?nèi)毖俟嘧⒔M明顯減少(p如.05)。2、電鏡結(jié)果顯示缺血/再灌注+丙泊酚組細(xì)胞凋亡不顯著,
