1、纖維連結(jié)蛋白在腎間質(zhì)纖維化大鼠腎組織中的表達程紅新1楊曉萍1蔣雅紅3林智峰1趙瑾2陶林2新疆石河子大學(xué)醫(yī)學(xué)院,1第一附屬醫(yī)院腎病科,2病理科,3第三附屬醫(yī)院醫(yī)務(wù)部,新疆維吾爾自治區(qū)石河子市832008摘要:背景:纖維連結(jié)蛋白(Fibronectin,F(xiàn)n)表達增加在腎間質(zhì)纖維化疾病進展過程中發(fā)揮著的重要作用。目的:探討纖維連結(jié)蛋白在腎間質(zhì)纖維化(RIF)大鼠腎組織中的表達及作用。方法:72只SD大鼠隨機摸球法均分為正常組、對照組和模型組。造模組建立單側(cè)輸尿管梗阻模型;對照組僅進行假手術(shù);正常組不做任何處理。分別于造模后第1,3,7,1
2、4天分批處死大鼠,檢測大鼠梗阻側(cè)腎小管間質(zhì)纖維化損傷程度并檢測腎臟組織中纖維連接蛋白(FN)、基質(zhì)金屬蛋白酶-9(MMP-9)、轉(zhuǎn)化生長因子-β1(TGF-β1)的表達。結(jié)果與結(jié)論:與正常組及對照組比較,梗阻時間越長,腎小管間質(zhì)損害程度越重,纖維化越明顯。造模后1d大鼠腎臟組織FN、MMP-9表達水平即出現(xiàn)上調(diào),造模后3d,TGF-β1表達水平出現(xiàn)上調(diào),并均于第14天達最高水平,與同期正常組、對照組相比,差異有顯著性意義(P<0.05)。說明在大鼠UUO腎纖維化形成過程中,F(xiàn)N的蛋白表達在腎損傷早期即顯著增加,并隨腎間質(zhì)纖維化程
