1、烏司他丁對(duì)膿毒癥大鼠肺微血管通透性及肺組織PEDF表達(dá)的影響作者:蔡施霞,鄧烈華,佟琳,孫小聰【摘要】目的觀察烏司他丁注射液對(duì)膿毒癥大鼠肺微血管通透性及肺組織色素上皮衍生因子(PEDF)表達(dá)的影響,探討烏司他丁對(duì)膿毒癥大鼠肺保護(hù)的作用機(jī)制。方法30只健康的雄性SD大鼠隨機(jī)分為3組:假手術(shù)組(A組)、陽(yáng)性對(duì)照組(B組)、烏司他丁治療組(C組),每組10例。除假手術(shù)組外,其他組均施行盲腸結(jié)扎穿孔術(shù)(CLP)復(fù)制大鼠膿毒癥模型,治療組同時(shí)給予烏司他丁腹腔注射(100ku/kg),術(shù)后4h隨機(jī)處死大鼠5只
2、取組織標(biāo)本,剩余5只大鼠經(jīng)頸外靜脈注射伊文思藍(lán)觀察肺微血管通透性。分別采用逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(RT-PCR)法和酶聯(lián)免疫吸附(ELISA)法檢測(cè)肺組織中PEDF的表達(dá)。結(jié)果B組肺微血管通透性明顯高于A、C組,C組肺微血管通透性高于A組;B組肺組織PEDF表達(dá)水平明顯低于A、C組,C組肺組織PEDF表達(dá)水平低于A組(P<0.05)。結(jié)論烏司他丁能明顯抑制膿毒癥大鼠肺微血管通透性增高,這種肺保護(hù)作用的機(jī)制可能與增加PEDF表達(dá)有關(guān)?!娟P(guān)鍵詞】烏司他丁注射液;膿毒癥;肺微血管通透性;PEDF 【

8、ammationresponsesyndrome,SIRS),進(jìn)一步發(fā)展可導(dǎo)致多器官功能障礙綜合征(MODS),其中呼吸功能障礙發(fā)生率較高,出現(xiàn)時(shí)間早,其主要的病理生理改變?yōu)榉蚊?xì)血管通透性增加。而色素上皮衍生因子(PEDF)可使血管通透性降低,雖然臨床研究證實(shí)烏司他丁有明確的肺保護(hù)作用,但其作用機(jī)制仍有待研究,本研究采用盲腸結(jié)扎穿孔術(shù)(CLP)復(fù)制大鼠膿毒癥模型觀察烏司他丁對(duì)膿毒癥大鼠肺微血管通透性和PEDF影響,探討烏司他丁對(duì)膿毒癥大鼠肺微血管通透性影響及其作用機(jī)制?! ?材料
