1、無色書虱AChE基因克隆及其系統(tǒng)進(jìn)化分析豆威1,唐培安1”,蔣紅波1,王進(jìn)軍1(1.西南大學(xué)植物保護(hù)學(xué)院,昆蟲學(xué)及害蟲控制T程重慶市市級(jí)霞點(diǎn)實(shí)驗(yàn)室,重慶400715;2.南京財(cái)經(jīng)大學(xué)食品科學(xué)與工程學(xué)院,南京210003)摘要:利用RT—PCR結(jié)合RACE技術(shù)克隆獲得了無色書虱尉acel的3’端序列和Ldace2的全長序列(GenBank登錄號(hào)分別為:F1647186和F]647187)。其中Ld∞e2基因全長2111bp,ORF1917bp,編碼638個(gè)氨摹酸組成的前體蛋白,成熟蛋白的分子量為71.9kDa,理論等電點(diǎn)為4.70,序列比對(duì)表
2、明該基因包括AChE家族的所有保守性功能位點(diǎn);刖acei基因僅獲得了3’端1616bp的序列,編碼500個(gè)氨基酸殘基,序列比對(duì)發(fā)現(xiàn),除缺少形成一對(duì)二硫鍵的半胱氨酸外,從acel包含了AChE家族的所有保守性功能位點(diǎn)。結(jié)合其它物種AChE構(gòu)建的分子進(jìn)化樹可以看出,AChE分為4個(gè)類群:昆蟲I型AChE類群、脊椎動(dòng)物AChE類群、昆蟲Ⅱ型AChE類群和線蟲AChE類群,其中無色書虱尉acel和蹦ace2分別屬于昆蟲I型AChE類群和昆蟲Ⅱ型AChE類群,且與嗜卷書虱和嗜蟲書虱的AChE具有最近的進(jìn)化關(guān)系。關(guān)鍵詞:無色書虱;乙酰膽堿酯酶;基因克隆
