1、神經(jīng)病理性疼痛大鼠行為學(xué)及生長(zhǎng)相關(guān)蛋白表達(dá)的變化厲彩霞林海王均爐△溫州醫(yī)學(xué)院附屬第一醫(yī)院麻醉科[摘要]目的:觀察大鼠坐骨神經(jīng)結(jié)扎后神經(jīng)病理性疼痛行為學(xué)以及生長(zhǎng)相關(guān)蛋白-43(GAP-43)表達(dá)的變化。方法:采用坐骨神經(jīng)結(jié)扎損傷(CCI)模型,75只SD大鼠,隨機(jī)分為三組(n=25)。對(duì)照組(con組):不接受任何手術(shù)操作;假手術(shù)組(sham組):只分離坐骨神經(jīng),不予以結(jié)扎;CCI組:結(jié)扎坐骨神經(jīng)。于術(shù)前2d,術(shù)后1、3、5、7、10、14、21、28d測(cè)定各組大鼠熱縮足反射潛伏期(TWL),機(jī)械縮足反應(yīng)閾值(MWT),并于術(shù)
2、后3、7、14、21、28天各組隨機(jī)抽取5只大鼠殺死,采用免疫組織化學(xué)方法測(cè)定脊髓背根節(jié)GAP-43表達(dá)的變化。結(jié)果:與sham組和比較,CCI組術(shù)后各日TWL,MWT明顯降低(P<0.01或<0.05),sham組和con組行為學(xué)無顯著差異(P>0.05)。CCI組較其它組GAP-43表達(dá)增高,且此蛋白在CCI組中術(shù)后第三天表達(dá)開始增高,第七至十四天達(dá)最高峰,以后逐漸下降。結(jié)論:CCI造模成功后大鼠痛覺及機(jī)械痛覺均過敏,且GAP-43表達(dá)增高。[關(guān)鍵詞]神經(jīng)病理性疼痛行為學(xué)改變GAP-43Behaviorlobservat
