1、實用標準文案自制纖維素金屬螯合親和膜的部分吸附性能及純化牛血SOD劉建榮1,張昊2,顧雅君1,趙曉瑜1(1.河北大學生物工程技術(shù)研究中心院,保定071002;2.河南雙匯投資發(fā)展股份有限公司,漯河462000)摘要:目的自制纖維素金屬螯合親和膜,研究親和膜對金屬離子(Cu2+、Ni2+、Fe3+)和蛋白質(zhì)的吸附性能,以求建立一種簡便實用的純化SOD的方法。方法以纖維素濾紙為原料,經(jīng)交聯(lián)制成可螯合金屬離子的纖維素親和膜,篩選確定與親和膜吸附量最大的金屬離子,并以卵清蛋白的吸附-脫附實驗考查該膜對蛋白質(zhì)的吸附性能。結(jié)果在Cu2
2、+、Ni2+和e3+中,親和膜對Cu2+的吸附量最大,多次重復實驗Cu2+吸附量穩(wěn)定維持在15.0μmol·(cm2)-1左右。Cu2+親和膜對生理鹽水溶卵清蛋白的吸附量高于無離子水溶卵清蛋白,但蛋白脫附率前者低于后者。吸附-脫附方式(靜態(tài)法)分離純化SOD,脫附樣品比活達8770U·mg-1,純度為81%;吸附-洗脫方式(動態(tài)法)分離純化SOD,穿柱流出樣品比活達12400U·mg-1,純度為85%。結(jié)論自制的纖維素親和膜有較好的吸附性能和純化效果,重復使用性能良好,制備和純化過程簡便易行。關(guān)鍵詞:親和膜;金屬螯合;纖維

7、igherthanthatdissolvedindeionizedwater,butasforproteinrecovery,theformerwaslowerthanthelatter.Thesamplewithspecificactivityof8770U·mg-1,purityof81%wasdesorbedbystaticadsorption-desorption.Theliquidthatpassedthroughcolumnbydynamicadsorption-elutionwasspecificactivi
8、tyof12400U·mg-1andpurityof85%.CONCLUSIONSelf-madecellulosemetal-chelatedaffinitymembranehasarathergoodabsorbingcharacteristicsandpurifyingeffect,adaptwe
