1、Notch信號通路相關(guān)分子在牙周膜干細(xì)胞中的表達(dá)及意義袁萍,李淑慧,于莉,趙璐,周春梅,吳佩玲*新疆醫(yī)科大學(xué)第二附屬醫(yī)院口腔科,新疆烏魯木齊市,830063[摘要]目的:檢測Notch信號通路相關(guān)分子在炎性和正常牙周膜干細(xì)胞中的表達(dá)差異,并探討其在牙周炎牙槽骨缺損中的作用機(jī)制。方法:組織快酶消化法培養(yǎng)正常及慢性牙周炎組織來源的的牙周膜干細(xì)胞(H-PDLSCs,P-PDLSCs),并經(jīng)有限稀釋法純化兩種細(xì)胞,采用免疫熒光法檢測干細(xì)胞表面標(biāo)記物CD146及PDLSCs表面蛋白STRO-1的表達(dá),并通過免疫熒光染色和實時熒光定量PCR檢測Notch信號
2、通路中Notch1受體蛋白、配體Jagged1(JAG1)信號分子的表達(dá)變化。結(jié)果:兩種來源的細(xì)胞經(jīng)純化后均陽性表達(dá)間充質(zhì)干細(xì)胞表面標(biāo)志物STRO-1、CD146;P-PDLSCs比H-PDLSCs具有更強(qiáng)的增殖能力;免疫熒光染色檢測顯示,H-PDLSCs和P-PDLSCs均陽性表達(dá)Notch1、Jagged1;但P-PDLSCs中Notch1、Jagged1的陽性表達(dá)明顯較弱;RT-PCR檢測發(fā)現(xiàn)P-PDLSCs中Notch1、Jagged1mRNA表達(dá)量分別為(1.22±0.29)、(1.52±0.38),較H-PDLSCs明顯降低,差異有統(tǒng)
