2、nstainingofhistologysectionofthegonadsandmacroscopicexaminationfistly,phenotypicsexofjuvenileindividualsfrom3to5monthsagewereidentified.Thensexratioof4022individualswithin37familiesat4monthswereanalyzedbothatfamilyandpopulationlevel.Meanwhile,2254individualsfrom20

3、familiesamong37familieswererandomlyselectedforanalysisofsexualdimorphisminbodyweight.CandidateSNPlocicomefromlarge-scalehigh-throughputsexspecialtranscriptome454sequencingsystemofturbot(ScophthalmusmaximusL.).AccordingtothecandidateSNPloci,primersweredesignedtotes

4、tandverify.AndbyuseofMassArray(MALDI-TOFMSArray),ahigh-throughputgenotypingtechnologyforSNPs,398sexassociatedcandidateSNPswerescreenedandverifiedwith20malesandfemalesat8monthsage.Thentheother35sex-associatedcandidateSNPsof6contigswereverifiedandconductedhaplotypes

5、analysis.Themainresultsareasfollows:1.Bydissectingindividualsat4monthsagedirectly,significantshapedifferenceofgonadsbetweenmaleandfemaleswereobserved.Females’wereshortrod-likewhilemalesaslenderline-like.Then,sexofmost4monthsageindividualscouldbeidentifiedbymacrosc

6、opicexamination.Fewindividualsthatshapeofgonadsshowednosignificantdifferencebetweensexes,stillshouldbeidentifiedbyuseofhaematoxylin-eosinstainingofhistologysectionoftheirgonads.Andashapedifferenceofgonadswasobservedbetween4and6monthsageindividuals,whichsuggestedth

7、atshapeofturbotgonadswascontinuouslychangingduringearlystage.2.Phenotypicsexof4022individualscomposedof37familiesat4monthsagewereidentifiedandcounted.Thestatisticalanalysisshowedthatthenumberoffemaleandmalewere2426and1596,respectively,whichreflectedasignificantdif
