1、江蘇醫(yī)藥2012年12月第38卷第23期JiangsuMedJ,December2012,Vol38,No.23·2787··論著·靜電紡絲PLGA納米纖維支架修復(fù)豬膽管損傷毛靜芳李佳許蓉蓉余澤前楊揚(yáng)周家華【摘要】目的觀察不同質(zhì)量比聚乳酸-羥基乙酸(PLGA)膽總管支架在體外膽汁中降解情況,實(shí)驗(yàn)探索可降解支架修復(fù)膽道損傷的可行性。方法用掃描電鏡觀察不同質(zhì)量比的PLGA納米纖維膜微觀形貌,并測定其短期體外膽汁中降解率。觀察其在膽總管Ⅰ期縫合、膽總管切開置入支架和膽管缺損修補(bǔ)手術(shù)中的作用。術(shù)后觀察動物黃疽和膽漏情況和動物自身術(shù)前和術(shù)后肝酶(ALT、AST
2、、ALP)和血膽紅素的變化。進(jìn)行Masson染色和平滑肌肌動蛋白(α-SMA)、增殖細(xì)胞核抗原(PCNA)免疫組化觀察肝臟及膽管損傷部位組織病理學(xué)改變。結(jié)果PLGA75/25和90/10兩種比例的纖維在膽汁中沒有發(fā)生明顯的降解和變形。膽管缺損修補(bǔ)手術(shù)的豬膠原增生程度輕,排列相對有序,α-SMA免疫組化評分最低,PCNA免疫組化示膽管缺損區(qū)域有新生膽管上皮細(xì)胞形成。結(jié)論可降解靜電紡絲PLGA納米纖維支架能有效支撐和修復(fù)膽管缺損?!娟P(guān)鍵詞】膽管損傷;靜電紡絲納米纖維;聚乳酸;羥基乙酸【中圖分類號】R364【文獻(xiàn)標(biāo)識碼】A【文章編號】0253-3685(2
