1、抗壞血酸誘導(dǎo)體外培養(yǎng)小鼠胚胎干細(xì)胞分化為心肌細(xì)胞李捷,張宜明,劉穎,夏暉,于長海(解放軍總醫(yī)院第一附屬醫(yī)院心胸外科,北京100037)提要:目的 探討采用抗壞血酸誘導(dǎo)體外培養(yǎng)小鼠胚胎干細(xì)胞(mouseembryonicstemcell,ES)分化為心肌細(xì)胞的可行性。方法未分化ES細(xì)胞在滋養(yǎng)層上長至匯合,胰酶消化成單個(gè)細(xì)胞經(jīng)旋轉(zhuǎn)生物反應(yīng)器制備擬胚體(embryoidbodies,EB),EB貼壁后用抗壞血酸誘導(dǎo)分化為心肌細(xì)胞。通過光鏡觀察跳動的心肌細(xì)胞,采用免疫組織化學(xué)、激光共聚焦和RT-PCR的方法鑒定心肌細(xì)胞。結(jié)果抗壞血酸能有效地誘導(dǎo)小
2、鼠胚胎干細(xì)胞向心肌細(xì)胞的分化,誘導(dǎo)效率為90%;分化的心肌細(xì)胞自發(fā)而有節(jié)律地收縮,心肌肌鈣蛋白T(cTnT)、α-輔肌動蛋白(a-actinin)、肌動蛋白(actin)表達(dá)陽性。此外,分化的心肌細(xì)胞還表達(dá)調(diào)控心肌發(fā)育的基因a-MHC、?-MHC。結(jié)論 抗壞血酸能有效地誘導(dǎo)體外培養(yǎng)小鼠胚胎干細(xì)胞分化為心肌細(xì)胞。關(guān)鍵詞:小鼠胚胎干細(xì)胞;心肌細(xì)胞;抗壞血酸;細(xì)胞培養(yǎng)中圖法分類號:文獻(xiàn)標(biāo)識碼:AAscorbicacidinducesdifferentiationofmouseembryonicstemcellsintocardiacmyocyt

7、embryonicstemcells;cardiomyocyte;ascorbicacid;cellculture獲得足夠數(shù)量、不引起免疫排斥反應(yīng)且具有再生活力的心肌細(xì)胞是開展組織工程研究的前提和基礎(chǔ)。胚胎干細(xì)胞(embryonicstemcell,ESC)是從早期哺乳動物囊胚的內(nèi)細(xì)胞團(tuán)發(fā)育而來,在體外具有無限增殖能力和發(fā)育的全能性。胚胎干細(xì)胞作為心肌組織工程的種子細(xì)胞來源,除需要對擬胚體(embryoidbodies,EB)進(jìn)行規(guī)?;瘮U(kuò)增以獲得足夠數(shù)量ES細(xì)胞此外,還需要將獲取的ES細(xì)胞在一定條件下,定向誘導(dǎo)分化為心肌細(xì)胞才有望用于心肌
